Callus induction and regeneration via shoot tips of Dendrocalamus hamiltonii
نویسندگان
چکیده
By using shoot tips as explants, various media and culture conditions for callus induction and proliferation, shoot differentiation, root induction and plantlet transplantation to develop an efficient and reliable regeneration system with Dendrocalamus hamiltonii were tested. Murashige and Skoog (MS) medium supplemented with 3 mg/l 2, 4-dichlorophenoxyacetic acid, 1 mg/l benzyladenine (BA), 500 mg/l glutamine, 500 mg/l proline, and 500 mg/l casein hydrolysate yielded the best rates of callus induction and granular-compact callus induction. MS medium supplemented with 1 mg/l BA, 0.3 mg/l kinetin and 0.3 mg/l naphthaleneacetic acid conferred the highest differentiation rate of calli. The maximum rooting rate was obtained in 1/2 MS medium supplemented with 3 mg/l indole-3-butyric acid, and the roots were long and thick. All hardened plantlets survived after transfer to an equal ratio mixture of peat, vermiculite and perlite. The regeneration system of D. hamiltonii developed is efficient and provides a useful tool for genetic transformation in bamboo species.

In vitro Plant Regeneration from the Callus of Shoot Tips in Cotton (Gossypium hirsutum L. cv. SVPR 2)
A high efficiency shoot organogenesis from callus cultures and plant establishment protocol has been developed for cotton (Gossypium hirsutum L. cv SVPR-2) through shoot tip. After 6 weeks of culture, green-compact-nodular organogenic callus induction was observed on the medium fortified with (MS) basal salts, B5 vitamins, 2.0 mgl-1 of benzylaminopurine (BAP), 1.0 mgl-1 kinetin (KN), 30 gl-1 gl...

The effects of plant growth regulators were examined in order to optimize the callus induction, regeneration, and proliferation of lisianthus (Eustoma grandiflorum). In vitro leaves provided the explants for callus induction. Explants were cultured on Murashige and Skoog (MS) medium with different concentrations of indole-3-acetic acid (IAA), 1-naphthaleneacetic acid (NAA), and 2,4-dichlorophen...
متن کاملAn Efficient Plant Regeneration and Transformation System of Ma Bamboo (Dendrocalamus latiflorus Munro) Started from Young Shoot as Explant
Genetic engineering technology has been successfully used in many plant species, but is limited in woody plants, especially in bamboos. Ma bamboo (Dendrocalamus latiflorus Munro) is one of the most important bamboo species in Asia, and its genetic improvement was largely restricted by the lack of an efficient regeneration and transformation method. Here we reported a plantlet regeneration and A...
متن کاملEfficient protocol for break impasses of regeneration via callus for 20 genotypes of chickpea
The results of studies on in vitro regeneration of chickpea (Cicer arietinum L.) have shown that indirect regeneration via callus was not successful. The introduction of an efficient and repeatable regeneration protocol is necessary for using transgenic technologies for the improvement of chickpea. The present study was undertaken to investigate the possibility of indirect regeneration in vario...
متن کاملMatthiola incana Micropropagation Using Shoot Tips and Callus Induction Derived from Lamina Explants and Rooting Capacity from Callus
Tissue culture is an attractive alternative for plant propagation. Micropropagation is a technique to ensure a constant and uniform source of ornamental plants. Matthiola incana is an important ornamental species mainly cultivate by seed. Matthiola incana seeds were germinated on solid MS medium without plant growth regulators. Shoot proliferation and root formation are possible using kinetin (...
